Andrographis Safety Issues
In general, Andrographis paniculata is considered as a safe and efficacious supplement for the relief of symptoms of
uncomplicated upper respiratory tract infection. However, there were also reports on its side effects.
Andrographis Side Effects
Andrographis side effects are generally mild and infrequent.[1-4] Its side effects may include gastric upset, headache,
bitter taste and fatique. Since it has effects on liver, high doses of Andrographis paniculata extracts may affect liver
In a study on its efficacy on common colds among 130 children, the Andrographis preparation was well tolerated and no
side effects or adverse reactions were reported in the study.
The dosages of Andrographis used in studies are summarized below:
The andrographis dosages used in a mice study on liver cancers are 50 and 100 mg/ kg body weight. Both
Andrographis treated groups showed a significant decrease in activity of LDH and malondialdehyde (MDA) formation,
and other biological alternations. These findings indicate the chemopreventive potential of Andrographis paniculata
against chemotoxicity including carcinogenicity.
Rats pretreated with andrographis dosage of 250 or 500 mg/kg showed significantly better inhibition of gastric
mucosal lesions.
The patients were randomized to receive either paracetamol or (dosage) 3 g/day of Andrographis paniculata or 6 g/day
(dosage) of Andrographis paniculata for 7 days. The baseline characteristics of the patients among the three groups
were not different. The efficacy of paracetamol or high dose Andrographis paniculata was significantly more than that of
low dose Andrographis paniculata at day 3 in terms of the relief of fever and sore throat.
Andrographolide at the effective dose (dosage 1.5 mg/kg) significantly attenuated the increase of plasma glucose
induced by an intravenous glucose challenge test in normal rats
HIV (Human Study)
The planned regimen (i.e. andrographolide from Andrographis paniculata Dosage) was 5 mg/kg bodyweight for 3
weeks, escalating to 10 mg/kg bodyweight for 3 weeks, and to 20 mg/kg bodyweight for a final 3 weeks. The trial was
interrupted at 6 weeks due to adverse events including an anaphylactic reaction in one patient
In the in vivo study, intra-peritoneal treatment of ovalbumin-immunized and nasally-challenged mice with
andrographolide significantly inhibited the elevation of bronchoalveolar fluid (BAF) levels of TNF-alpha and GM-CSF in
a dosage-dependent manner, with 30 mg/kg producing an inhibition of 92% and 65% of the cytokines, respectively)
and almost completely abolishing the accumulation of lymphocytes and eosinophils.
