Hydroxypropyl methylcellulose
Hypromellose has the polymeric backbone of cellulose, a natural carbohydrate
that is based on a repeating structure of anhydrous units. It is used as an
ophthalmic lubricant. As a pharmaceutical ingredient, it is used as emulsifier,
binder, spending and controlled release agents.
Uses
As a lubricant in ophthalmic product
Hypromellose is also applied topically to the conjunctiva to protect the cornea
during certain ophthalmic procedures and used as artificial tears to lubricate the
cornea. Side effects include blurred vision and stickiness of eyelashes. [1,2]
As a thickening agent in liquid product
High-molecular weight of hypromellose such as METHOCEL E4M and K4M Premium
grades are most frequently used to modify viscosity in many cream, ointment, and
suspension products.
As a binder in granules/tablets
Low-molecular weight hypromellose such as METHOCEL E5LV and E15LV can be
used as tablet binders in the wet granulation of tablet products at a level of 3-5%
of finished tablet weight. Use of METHOCEL™ E15LV may provide stronger
granules, but it may also slow down the disintegration of your tablet.
As a controlled-release agent in tablet and bead products
Hypromellose is often used for the formulation of hydrophilic matrix systems,
providing a mechanism for the slow release of drugs from oral solid dosage forms.
There are different grades of hypromellose, based on their viscosity. In general,
low molecular weight hypromelloses, such as METHOCEL E50LV, K100LV, K100LV
CR, is used to prepare hydrogel tablets for low solubility drugs while high
molecular weight hypromelloses, such as METHOCEL K4M, K15M, K100M, E4M,
E10M, K4MCR, K15MCR, K100MCR, E4MCR, E10MCR, are used in hydrogel tablets
for water-soluble drugs.
